The victim was fatally injured in a confrontation involving "multiple people" at the school in Wilmington, said the mayor's office.
Police said they were called to Howard High School of Technology at around 8.20am after a report of a serious assault.
According to 6ABC.com, the girl hit her head on a sink during the fracas and lost consciousness.
Student Kayla Wilson told the news station she was in a toilet cubicle when the brawl broke out.
"She was fighting a girl, and then that's when all these other girls started banking her, like jumping her, and she hit her head on the sink," she said.
The critically injured victim was flown by state police helicopter to hospital, where she was pronounced dead, police said.
Two female students were taken to police headquarters for questioning, police Chief Bobby Cummings told the News Journal.
Wilmington Mayor Dennis Williams told a news conference: "I'm so upset that the young lady lost her life today.
"Things like this shouldn't happen. My heart bleeds for the family, the kids that go to this school, administrators and our city."
Detectives from the homicide and violent crimes unit are working on the case.
According to the News Journal, a parent received a text from her daughter who is a student there saying "there was a riot at the school".
Classes at the school were cancelled for the rest of the day.
